Schiff Reagent

Solution

Ingredient	Amount
Pararosanilin	0.2 g
Distilled water	400 mL
Hydrochloric acid, 1N	4 mL
Sodium thiosulphate	96 g

 

Preparation
Dissolve the dye in boiling distilled water.
Add the sodium thiosulphate and mix well.
Filter.
Add the hydrochloric acid.
Stir until the solution is crystal clear and slightly pink (1-2 hours), with a pink to white precipitate.
Refrigerate overnight.
Store in a tightly stoppered bottle in the dark.
Filter before use.

 

Notes

1. The solution should be colourless or very pale amber. Brown solutions will often stain poorly. Usually these have been made from a batch of basic fuchsin containing too little pararosanilin.
